Post-translational regulation of DGAT1, futile lipid cycling and energy expenditure

Futile lipid cycling, driven by lipolysis and re-esterification of fatty acids, represents an alternative mechanism of thermogenesis in brown and beige adipose tissue. We previously demonstrated that this pathway is activated upon β-adrenergic stimulation in the absence of UCP1 and identified DGAT1 as a key regulator of the cycle. Our current work investigates how DGAT1 activity is regulated. Using cellular and in vivo models, stable isotope tracing, and direct and indirect calorimetry, we aim to define the contribution of DGAT1-regulated futile lipid cycling to UCP1-independent thermogenesis.
